Anticoagulants on yield of bone marrow-derived mononuclear cells harvested from dogs Ciência Rural
Brito,Harald Fernando Vicente de; Fernandes-Filho,Tobias; Brandão,Fernanda de Lima; Montaño,Patrícia Yukiko; Laskoski,Luciane Maria; Locatelli-Dittrich,Rosangela.
Cell therapy with bone marrow-derived mononuclear cells is an alternative to therapy with mesenchymal stem cell cultures. The aim of the present research was the comparison of the yield of bone marrow-derived mononuclear cells harvested from dogs with two different anticoagulants. Bone marrow was harvested from the iliac crest of five healthy dogs aged between 15 and 30 months, and the effect of two anticoagulant solutions, CPDA-1 (citrate phosphate dextrose adenine-1) and heparin, on the isolation of mononuclear cells was compared. Mononuclear cells were isolated in a density gradient and stained for CD9 and CD44 for characterization by flow cytometry. Means were compared using Student's paired t-test. Samples harvested with CPDA-1 yielded an average of...

Hematological and immunological development from birth to six months of age in Holstein calves Arq. Bras. Med. Vet. Zootec.
Baccili,C.C.; Oliveira,S.M.F.N. de; Costa,J.F.R; Massoco,C.O.; Pozzi,C.R.; Gomes,V..
ABSTRACT The hematological and immunological development of calves from birth to 6 months of age was performed by hemogram and cellular phenotype. Ten male Holstein calves were assessed in 13 moments: before colostrum intake (D0), every 2 days until the 10th day of life (D2 to D10), at the 15th day after birth (D15), and then monthly up to 6 months (D180). Calves presented hemoconcentration on day (D) 0 and showed a gradual decrease in hematimetric rates until D180. The inversion of the neutrophilic for lymphocytic profile was observed on D4. The percentage of CD3+ cells on D10 was higher than D30 up to D180. The number of CD4+ and CD8+ cells did not change between time points. The number of CD21+ lymphocytes was significantly higher at early time points...

Imbalance of naive and memory T lymphocytes with sustained high cellular activation during the first year of life from uninfected children born to HIV-1-infected mothers on HAART BJMBR
Ono,E.; Santos,A.M. Nunes dos; Succi,R.C. de Menezes; Machado,D.M.; Angelis,D.S.A. de; Salomão,R.; Kallás,E.G.; Moraes-Pinto,M.I. de.
The immune consequences of in utero HIV exposure to uninfected children whose mothers were submitted to highly active antiretroviral therapy (HAART) during gestation are not well defined. We evaluated 45 HIV-exposed uninfected (ENI) neonates and 45 healthy unexposed control (CT) neonates. All HIV-infected mothers received HAART during pregnancy, and the viral load at delivery was <50 copies/mL for 56.8%. Twenty-three ENI neonates were further evaluated after 12 months and compared to 23 unexposed healthy age-matched infants. Immunophenotyping was performed by flow cytometry in cord and peripheral blood. Cord blood lymphocyte numbers did not differ between groups. However, ENI neonates had a lower percentage of naive T cells than CT neonates (CD4+, 76.6...

Immunomodulatory activity of Tityus serrulatus scorpion venom on human T lymphocytes J. Venom. Anim. Toxins incl. Trop. Dis.
Casella-Martins,Andrea; Ayres,Lorena R; Burin,Sandra M; Morais,Fabiana R; Pereira,Juliana C; Faccioli,Lucia H; Sampaio,Suely V; Arantes,Eliane C; Castro,Fabiola A; Pereira-Crott,Luciana S.
Abstract Background Tityus serrulatus scorpion venom (TsV) contains toxins that act on K + and Na + channels and account for the venom’s toxic effects. TsV can activate murine peritoneal macrophages, but its effects on human lymphocytes have been poorly investigated. Considering that lymphocytes may play an important role in envenomation, we assessed whether TsV affects the expression of phenotypic (CD3, CD4, and CD8) and activation (CD69, CD25, and HLA-DR) markers, cell proliferation, and cytokine production in peripheral blood mononuclear cells. Methods Cytotoxicity of TsV was evaluated via the MTT assay. Cell proliferation, expression of phenotypic and activation markers, and release of cytokines were assessed using flow cytometry, after treatment...

Immunophenotypic evaluation, and physiological and laboratory correlations of hematopoietic stem cells from umbilical cord blood Biocell
Canabarro,Raquel; Sporleder,Heloísa; Gomes,Thaís; Zanatta,Geancarlo; Scribel,Letícia; Freitas,Fernando; Neumann,Jorge; Pranke,Patricia.
The use of umbilical cord blood stem cells is an efficient alternative for the transplantation of hematopoietic progenitor cells. A number of factors can influence the volume and amount of CD34+ cells, which are considered as immature and capable of proliferation. Quantification of CD34+ cells, evaluation of CD38 and c-kit molecules on these cells, as well as correlations of such factors as maternal age, gestational age, newborn sex and weight, umbilical cord length, placental weight with increased volume and concentration of immature cells, among others, were performed in 70 blood samples from term newborns. The mean volume of umbilical cord blood collected was 53.8±33.6 mL, where 30.96±18.9 CD34+/mL UCB cells were found, of which 16.66±8.32% were...

T lymphocyte subsets of the umbilical cord blood of dogs Arq. Bras. Med. Vet. Zootec.
Cápua,M.L.B.; Santana,A.E.; Nakage,A.P.M.; Godoy,A.V.; Kataoka,A..
The hematological parameters red blood cells (RBC) and total white blood cells (WBC) counts, hematocrit, hemoglobin concentration, and RBC indexes (median corpuscular volume and median corpuscular hemoglobin concentration) were determined and T CD5+ lymphocytes and CD4+ and CD8+ subpopulations of the umbilical cord blood (UCB) of dogs were quantified by the cytofluorimetric technique. Nine adult Beagles, from two do five-year old, were used as control. The umbilical cord blood (UCB) was collected from 20 neonate dogs. The method for the UCB collection was adequate to obtain sufficient quantity of blood for the accomplishment of the hematological analyses and lymphocyte quantification. Cytoscopic preparations of the UCB suggested high erythropoietic...
